Intended use:

 

This kit is used for the specific detection of the Tilapia Lake virus (TiLV) in liver, spleen, kidney, and brain tissue samples of silk perch, Chinese mitten carp, Chinese giant salamander, and zebrafish.
 
 

Principle:

 

The kit synthesizes cDNA strands complementary to the RNA template using RNA as the template and primers as the starting point under the action of efficient reverse transcriptase. The specific primers and specific fluorescent probes are designed for the nucleic acid conserved region of Tilapia Lake Virus.

 

The Real Time PCR reaction solution and real-time fluorescent quantitative PCR detection technology is applied on the fluorescence quantitative PCR instrument for realizes the rapid detection of Tilapia Lake Virus through the change of fluorescence signal.

Sample requirements:
 

1. Sample type

Adult shrimp, shrimp, shrimp seedlings, bait, processed aquatic products, etc.

2. Transport

Sample collection and transportation shall be carried out in accordance with NY/T 541. Samples shall be transported to the laboratory as soon as possible under refrigerated conditions to avoid repeated freeze-thaw.

3. Sample preservation

It can be stored for 24 hours at 2℃~8℃, and can be stored for a long time at -20℃.

 
PCR amplification and fluorescence Detection (PCR detection area)

Put each reaction tube into the PCR instrument and perform PCR amplification according to the following procedures:

Note: The result is judged to be grey area, which needs to be rechecked. If the recheck result is Ct value < 45 is considered as positive, and no Ct value or Ct value ≥45 is considered as negative.
